Three-dimensional mesenchymal stem cell laden scaffold of icariin sustained-release for bone regeneration

Background/aim: Icariin (ICA)-loaded zein/PLGA nanofiber membrane combined with MSCs was prepared by coaxial electrospinning and bioelectrospraying. Materials methods: SEM TEM were used to evaluate the surface morphology microstructure of fiber membrane. Ultraviolet spectrophotometry detect drug release. A LIVE/DEAD Viability/Cytotoxicity Kit fluorescence staining cell activity. Alkaline phosphatase calcium mineralization deposition osteoinductive activity scaffold. Dynamic mechanical analysis determine Young's modulus, maximum load, elongation Western blot related protein expression in induced drug-loaded scaffolds. Results: Good properties stability observed showed that there a considerable number dispersed evenly distributed, could grow between fibers, arranged orderly along fibers. Sustained release ICA confers laden scaffold higher ALP ECM mineral through Runx2, OPN OCN pathways. Conclusion: Isotropic sustained grant scaffolds rapid bone regeneration compacity which can provide good osteogenic environment for loaded MSCs.
